Cas9 Nuclease is an RNA-guided endonuclease that catalyzes site-specific cleavage of double stranded DNA. It is widely used to induce site-directed double strand breaks in DNA, which can lead to gene knock-out or gene knock-in through NHEJ and HDR respectively
ApplicationUsed for 1) In vitro DNA cleavage by gRNA guiding; 2) Genome editing through the injection for mammalian zygote, zebrafish embryos, & drosophila embryos; 3) Introduction into cell by transfection. In this buffer, Cas9 can directly be used for microinjection, eletroporation and in vitro DNA cleavage.

Physical AppearanceIn the buffer [20 mM Hepes pH 7.5, 150 mM KCl, 1 mM MgCl2, 5%glycerol, 1 mM Tris(2-chloroethyl) phosphate (TCEP)]. 0.2μm filter to sterilize.
FormulationThe protein concentrations can be at 5mg/ml (equal to 31μM), or at 1mg/ml (equal to 6.3μM) in a micro-injection buffer.
SolubilityThe Cas9 is very soluble in water and most aqueous buffers.
StabilityAlthough stable at room temperature, Cas9 should be stored desiccated below 0°C.

Amino acid sequenceThe N terminus contains a 6xHis-tag and the C-terminus contains a TAT peptide for the nuclear localization signal and cell-penetrating peptide(CPP).
Biological ActivityBiological activity is assayed by in vitro gRNA guide DNA cleavage.
Protein contentProtein purity of 98%. Quantification was carried out by Bradford methods.
